Damage Per Second.

Often used casually like "DPS champion" meaning a champion who's primary role is to deal a lot of consistent damage (aka AD auto attack carries). Anyone with high damage may be refereed to as a DPS champion, though champions with high spikes of burst damage and then long downtime while they wait for cooldows to come back are generally called "burst champions" instead.

DPS stands for Damage per second.
Generally means how much damage you deal in a short period of time.

I.E: Vayne has the highest DPS of all AD carries lategame.

Yes, it stands for damage per second. It can mean a character role. Carries for example, should have high DPS, so they are called a DPS class by some. In MMOs, dedicated damage dealers are usually called DPS. Their role is mainly to deal damage. For example, an assassin belongs to the DPS class, while a warrior could be a tank.

It usually refers to sustained damage.

It can also be an exact number. For example, if you have 200 AD and you attack twice each second (2 attack speed), your DPS is going to be 400, because you deal 400 damage per second. Of course things like crit chance, crit damage, attack speed armorpen, debuffs and enemy defenses affect DPS.
